I am investigating the mechanism of how the ER stress initiates and/or progresses motor neuron diseases, including ALS and Seipinopathy. Previously I revealed that aggregates of the ER membrane protein ngSeipin, a causal mutant protein of Seipinopathy, incorporate and inactivate ER calcium ion pomp SERCA2b, thereby inducing ER stress and cell death (Saito et al., Elife, 2022). And in my recent study, I found that ATF6-branch, one pathway of the Unfolded Protein Response (UPR), protects cell from ngSeipin not only by transcriptional induction of BiP, one of the most important ER chaperones, but also by optimization of its availability. In this forum, I would like to show you the new mechanism of how some of ATF6-target proteins realize optimal anti-aggregation effect of BiP.
